Run a 1H 1D spectrum with presaturation of the water siganal
1. Follow the instructions in
Run a 1D spectrum and calibrate 90° pulse width
2. Create a new experiment
Type edc to create a new experiment. Enter desired values for NAME and EXPNO. Set USER to your user name and DU=u, PROCNO=1, TYPE=nmr.
Type rpar Stan1Dps to load a standard parameter set. Press COPY ALL. Type ii to initialize the instrument.
3. Acquire and process a spectrum
Type zg to acquire an FID. Wait for the checklockshift:finished message.
Type ef to perform exponential multiplication and Fourier transformation.
Type apk to automatically phase the spectrum.
Note: more detailed instructions are available in Process a 1D spectrum.
4. Optimize parameters
Adjust the sweep width to accomodate the chemical shift range of your sample by typing sw and entering a new value in ppm.
If the signal-to-noise ratio is too low increase the number of scans by typing ns and enter a new value, which should be a multiple of 8.
If the spectrum contains spikes or the baseline is wavey the receiver gain may be too high. Type rg and enter a smaller value.
Optional: you can automatically optimize the receiver gain by typing rga.
Note: if you change rg you will have to rephase the spectrum.
Reacquire and process a new spectrum by typing zg, then efp.
