Protein-optimized 1H-15N HSQC

Description 2D 1H-15N HSQC, WATERGATE incorporated into final INEPT module, H2O magnetization returned to equilibrium prior to acquisition, magic angle gradients.

Instructions

  1. Create a 2D experiment and read in parameter set.

  2. Tune the probe. Go to the acqu window and start wobb. Tune the 15N channel. Push the Channel Selector button on the preamplifier, then tune the 1H channel.

  3. Create a 1D 1H experiment and acquire a 1D data set. Calibrate p1 (1H 90° pulse at pl1=0, 9 to 13 µs).

  4. Return to the 2D experiment. Set p1. Adjust ns as required for your sample. The default value is good for a 1 mM sample.

  5. In gs mode adjust sp1 and phcor14 iteratively until WATERGATE is optimized.

  6. Acquire a spectrum. Aliased peaks will have negative phase. Adjust 1 SW if needed.
